Parasites with complex life cycles often manipulate the behavior, morphology, physiology and life history of their intermediate hosts to increase transmission to their definitive host. Manipulations induced by parasites can vary from minor changes in the host phenotype to displays of entirely new behaviors, which can be regarded as the extended phenotype of the parasite. Despite the prevalence of parasites, which are estimated to make up more than half of all extant organisms, we are only now beginning to understand how parasites manipulate hosts on a molecular level. Some of the most dynamic examples of these manipulations involve social insects as hosts including our focal host-parasite system between the common endoparasitic tapeworm Anomotaenia brevis and its intermediate host, the ant Temnothorax nylanderi. The infected adult ants are easily identified by their yellow-soft cuticle. Infection lead in workers to an increased reproductive potential and lifespan whereas their nestmate workers suffer from a higher mortality. It remains unclear as to how long infected workers can slow aging through cestode infection.

The question arising from the tapeworms’ impact on host survival is how aging is modulated in these social insects on a proximate level. A well-documented hallmark of aging in Drosophila is the increased expression of genes involved in stress and immune response as well as expression of antimicrobial peptides. In our ant-cestode system, an increased lifespan of infected hosts compared to unparasitized nestmates could point to an interplay between immunity and lifespan. We aim to gain insights into the interplay and regulation of immunity and lifespan in insects by investigating the Anomotaenia brevis-Temnothorax nylanderi system. In particular, we question how does the cestode regulate and control immunity, aging and longevity of its ant host on a molecular level. We predict that the chronic immune activity of parasitized ants will be kept at bay by the Anomotaenia cestode causes the lifespan extension in the host. Also, due to the prolonged nature of the ant-cestode interaction (i.e. years), the cestode essentially inherits the parasites and enemies of its host. To evaluate the molecular basis of the host immune response, expression of genes involved in core immune pathways, will be detected and quantified using RNA-seq for selected time-points during various adult stages (from young to old) in parasitized compared to unparasitized ants.
